miR-708与JAK1在大鼠脑缺血再灌注损伤中的表达及调控关系Expression and interaction of miR-708 and JAK1 in rats with cerebral ischemia-reperfusion injury

摘要(Abstract):
目的初步探讨miR-708在大鼠脑缺血再灌注损伤中的作用及机制。方法选取25只雄性SD大鼠,随机抽取10只作为假手术组,其余大鼠采用线栓法构建大鼠脑缺血再灌注模型(MACO),将术后存活的10只SD大鼠设为缺血再灌注1d组,采用实时定量荧光PCR法(real time,PCR)比较各组大鼠外周血和脑组织的miR-708表达及脑组织中JAK1 mRNA的表达;蛋白免疫印迹法(Western blot)检测各组大鼠脑组织中JAK1蛋白的表达水平。结果与假手术组相比,实验组外周血及脑组织中miR-708mRNA表达显著降低(P<0.01),而脑组织中JAK1 mRNA表达水平明显上调(P<0.01)。相关性分析发现miR-708与JAK1表达呈负相关。结论 miR-708可能通过抑制其靶基因JAK1的表达在脑缺血再灌注损伤中发挥保护作用。
