UPS抑制剂对成年大鼠脑内淀粉样蛋白沉积和神经新生的影响

摘要(Abstract):
目的观察UPS抑制剂对成年大鼠脑内淀粉样蛋白沉积和神经前体细胞增殖水平的影响。方法采用水迷宫方法检测大鼠行为学习记忆能力,在腹腔注射UPS抑制剂60d后,采用水迷宫再次测定大鼠学习记忆能力,用免疫组化方法标记神经细胞增殖的方法,用改进的特殊刚果红染色方法观察大鼠脑内淀粉样蛋白沉积的变化。结果腹腔注射UPS抑制剂60d后,实验组与对照组相比,学习记忆能力和Brdu阳性细胞数量显著减推(P<0.05),而淀粉样蛋白沉积数量明显增加。结论 UPS抑制剂可能是通过影响了脑内淀粉样蛋白的沉积,减少了神经前体细胞的增殖而影响了成年大鼠的学习记忆能力。
