脑缺血后神经调剂素-1对缺血边缘区细胞凋亡的影响及机制

摘要(Abstract):
目的研究脑缺血后神经调剂素-1(NRG-1)对缺血边缘区细胞凋亡的影响及机制。方法制作大鼠颈内动脉栓塞再灌注模型,栓塞前经颈动脉注射NRG-1或生理盐水,缺血1 h再灌注23 h后,通过TUNEL法标记缺血皮层细胞凋亡,逆转录PCR观察Bcl-2 mRNA的表达,western bolt检测caspase-3的表达。结果与生理盐水对照组相比,NRG-1显著减少了缺血皮层的凋亡细胞比例,神经元凋亡减少亦明显。NRG-1显著上调Bcl-2 mRNA的表达,减少了caspase-3蛋白的表达。结论 NRG-1作为脑缺血后的一种重要保护因子,通过经典途径减少神经细胞的凋亡,从而起到脑保护作用。
