ApoE基因敲除小鼠主动脉组织中CD55、CD59表达的变化及意义The levels of complement regulatory proteins CD55 and CD59 expression on aortas in ApoE-knock out mice

摘要(Abstract):
目的探讨ApoE基因敲除小鼠主动脉组织中CD55、CD59的表达变化及意义。方法实验动物分为2组:对照组(C57BL/6小鼠10只)和动脉粥样硬化(AS)组(ApoE基因敲除小10只)。普通饲料喂养8周。监测体质量、血脂,观察斑块病变情况;采用免疫荧光和Real-ti me PCR技术,从蛋白水平和mRNA水平来观察CD55和CD59的表达变化。结果与对照组相比,AS组体质量增加[(27.14±1.60)g vs(29.50±1.38)g,P<0.05],总胆固醇和低密度脂蛋白明显增高〔(2.52±0.16)mmol/Lvs(12.74±1.79)mmol/L,(0.28±0.03)mmol/Lvs(9.06±1.13)mmol/L,P均<0.01〕;小鼠血清淀粉样蛋白-a水平有增加趋势[(5.08±2.34)ug/mLvs(7.06±4.81)ug/mL,P>0.05];AS组可见明显动脉粥样硬化斑块形成。免疫荧光显示,AS组CD55和CD59表达增强。Real-ti me PCR结果显示,与对照组比较,AS组CD55和CD59 mRNA相对表达量均有上升趋势,但差异无统计学意义(0.75±0.19 vs 4.20±0.45,1.21±0.21 vs 5.58±2.53,P均>0.05)。结论 ApoE基因敲除小鼠主动脉组织中CD55、CD59蛋白水平和mRNA水平表达上调,可能参与了动脉粥样硬化的发生和发展。
