重组人促红细胞生成素对氧化损伤的血管内皮细胞的保护作用

摘要(Abstract):
目的观察重组人促红细胞生成素对氧化损伤的血管内皮细胞的保护作用以及其机制。方法体外培养人脐静脉内皮细胞建立过氧化氢诱导的内皮细胞氧化应激损伤模型,不同重组人促红细胞生成素预处理组,用噻唑蓝(MTT)比色检测细胞活力,黄嘌呤氧化法和硫代巴比妥法分别测定细胞上清的超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量,流式细胞仪检测细胞凋亡。结果 H2O2对人脐静脉内皮细胞有损伤作用。rhEPO可增加细胞活性,抑制H2O2诱导人脐静脉内皮细胞的MDA的产生,增加SOD的活力及抑制细胞凋亡。结论重组人促红细胞生成素对氧化损伤的血管内皮细胞具有保护作用,其作用机制可能与抗氧化有关。
